How & where can I buy brake pads without taking them off first?

I'm pretty sure the 250 and 330 brakes used the same pads that were made by Dunlop. The usual Ferrari parts suppliers should be able to help. Jaguars used these pads as well, so there are other suppliers that can help. I recently saw a shop that even makes billet stainless steel pistons that will fit our Ferraris!

My 330 GT brakes are Dunlop, front 2 1/8", rear 1 5/8"  Go to the ssbrakes.com site in the restoration and import sections, and match up what is on your car (and note the other marques).  This could help sourcing across brands.  SSbrakes sells pads, also try JCWhitney for street, http://www.porterfield-brakes.com/ for the track.

BUT, in the long run, its a good idea to support your local ferrari supplier too, for when you need that part made of unobtaineum.
